Summary Introduction: Bioengineered human cardiac tissue has been developed worldwide as a tissue model. However, there are few reports on the development of aligned cardiac tissue model. Methods and Results: We created the micro V shaped surface with the height of 10µm and the interval of 30µm on silicon resin base materials. Molding fibrin gel on the V shaped silicon resin, the micro inversed V shaped surface were transcribed on the fibrin gel. After seeding human iPS cell-derived cardiomyocytes (hiPS-CM) on inversed V shaped fibrin gel (VFG), hiPS-CM aligned on VFG compared with hiPS-CM on normal fibrin gel (NFG) shown in upper Figure. Further, the directionality histogram showed each angle of actin filaments in hiPS-CM on VFG were more oriented in the same direction compared with those on NFG shown in lower Figure. Conclusions: Using this strategy, hiPS-CM might be oriented on fibrin gel. It might lead to develop more matured cardiac tissue model to investigate the contractile properties for disease research and drug development.
