| 概要 | To enable rapid cell dissociation for intraoperative brain tumor diagnosis, we developed a novel cell isolation device that generates shear stress between two high-speed rotating rollers. This study investigated the optimal roller gap to maximize dissociation efficiency, achieving single-cell suspensions from porcine brain tissue in approximately 30 s. Flow cytometry analysis revealed a maximum single-cell ratio of 98.47 士 0.72% at a peripheral roller velocity of 1.0 m/s with a 0.2 mm gap. These results define the optimal mechanical conditions for the device and support its utility in accelerating intraoperativediagnostic workflows.
