概要 | Methods for differentiating stem cells into cardiomyocytes by growth factors have been routinely used. Recently, the method using hybrid gels composed of native heart matrix and collagen has been reported to induce cardiac differentiation of human embryonic stem cells without supplemental growth factors. However, the influence of mechanical properties of the gel has not been thoroughly investigated since mechanical properties of scaffold is one of the critical factors for differentiation. In this study, we fabricate bio-hydrogel composed of goat cardiac extracellular matrix, and investigated the mechanical properties by means of uniaxial compression test. It showed that cardiac ECM gels from goat hearts possess viscoelastic property. The elastic modulus K1 in Kelvin model is at 6.3~18.6 Pa for these gels and K2 is at 29.6~131.7 Pa, consistently larger than K1. The relaxation coefficient λ is ca. 20 s. Moreover, this study found that the elastic modulus K2 remarkably increased by fixation. |
